Sequencing DNA
Using the dideoxy method of DNA sequencing, a single-stranded recombinant vector of DNA is synthesized. To determine the order of nucleotides in the newly made strand, select a nucleotide base, one at a time, and move it to the correct location in the gel.

Prior to DNA sequencing via the dideoxy method, a segment of DNA is cloned in a vector next to a primer annealing site. Many copies of the recombinant vector, primer, dNTPs, fluorescently labeled dideoxynucleotides, and DNA polymerase are mixed together and DNA synthesis occurs. Newly made strands are then separated by gel electrophoresis. This exercise contains a gel with empty spaces where bands would be located. Based on the DNA sequence in the recombinant vector on the left, drag the correct types of bands into the 14 empty spaces. When you are finished, click Submit.
